چکیده

A deformable mirror is used to scan the focal plane during the camera exposure, obtaining extended depth-of-field. A deconvolution kernel is approximated for a given scan depth and used to de-blur the image. OCIS codes: (180.2520) Fluorescence microscopy; (110.1080) Active or adaptive optics; (100.1830) Deconvolution

Extended depth-of-field microscopy with a high-speed deformable mirror.

We present a wide-field fluorescence microscopy add-on that provides a fast, light-efficient extended depth-of-field (EDOF) using a deformable mirror with an update rate of 20 kHz. Out-of-focus contributions in the raw EDOF images are suppressed with a deconvolution algorithm derived directly from the microscope 3D optical transfer function. Demonstrations of the benefits of EDOF microscopy are...

High-resolution wide-field microscopy with adaptive optics for spherical aberration correction and motionless focusing.

Live imaging in cell biology requires three-dimensional data acquisition with the best resolution and signal-to-noise ratio possible. Depth aberrations are a major source of image degradation in three-dimensional microscopy, causing a significant loss of resolution and intensity deep into the sample. These aberrations occur because of the mismatch between the sample refractive index and the imm...

Adaptive correction of depth-induced aberrations in multiphoton scanning microscopy using a deformable mirror.

We demonstrate adaptive aberration correction for depth-induced spherical aberration in a multiphoton scanning microscope with a micromachined deformable mirror. Correction was made using a genetic learning algorithm with two-photon fluorescence intensity feedback to determine the desired shape for an adaptive mirror. For a 40x/0.6 NA long working distance objective, the axial scanning range wa...

Image-based calibration of a deformable mirror in wide-field microscopy.

Optical aberrations limit resolution in biological tissues, and their influence is particularly large for promising techniques such as light-sheet microscopy. In principle, image quality might be improved by adaptive optics (AO), in which aberrations are corrected by using a deformable mirror (DM). To implement AO in microscopy, one requires a method to measure wavefront aberrations, but the mo...

A Novel High-Performance Field-Weakening Control for Axial Flux-Switching Permanent-Magnet Motor

By combining the field-weakening control principle of a new axial flux-switching permanent-magnet motor (AFFSSPM) with the space vector pulse width modulation (SVPWM) and maximum torque per voltage (MTPV) control principle, a novel field-weakening control strategy for AFFSSPM is proposed in this paper. In the first stage of the field-weakening, the difference between the reference voltage updat...
